The invention discloses silk fibroin nano-microspheres as well as a preparation method and application thereof, and belongs to the technical field of cosmetics. The silk fibroin nano-microsphere provided by the invention is of a core-shell structure, a core layer comprises cosmetic polypeptide, a shell layer comprises silk fibroin and ionic liquid, and the D50 particle size of the silk fibroin nano-microsphere is 1-500nm; the ionic liquid is a mixture formed by a quaternary amine alkali substance and at least one of a malic acid substance, a glycerol substance and a hydantoin derivative. The silk fibroin nano-microsphere provided by the invention has the characteristics of excellent biocompatibility, capability of effectively promoting cell proliferation and migration and capability of promoting secretion of cell collagen, and plays a slow release role while promoting transdermal absorption of effective components, so that the silk fibroin nano-microsphere is applied to the field of cosmetics, and the efficacy of the cosmetics is improved..
