The invention provides a biological switchable miR nano-composite based on DNA tetrahedral framework nucleic acid as well as a preparation method and application of the biological switchable miR nano-composite, and belongs to the field of nucleic acid molecule medicines. The nanocomposite is a tetrahedral framework nucleic acid obtained by complementary base pairing of three oligonucleotide chains and three micro RNAs, wherein the tail ends of the micro RNAs are connected with restriction enzyme cutting sites; each oligonucleotide chain is composed of two DNA (deoxyribonucleic acid) chains and two RNA (ribonucleic acid) chains according to a sequence of RNA1-DNA1-RNA2-DNA2; the DNA1 chains of the three oligonucleotide chains form three edges of the tetrahedral framework nucleic acid through complementary pairing of bases; the three microRNAs of which the tail ends are connected with restriction enzyme cutting sites are respectively paired with the RNA1 chain, the RNA2 chain and the DNA2 chain of the oligonucleotide through base complementation to form the other three edges of the tetrahedral framework nucleic acid, each micro RNA is respectively paired with the RNA2 chain of one oligonucleotide chain and the RNA1 chain of another adjacent oligonucleotide chain through base complementation, and the restriction enzyme cutting sites connected with the tail ends of the microRNAs are paired with the DNA2 chain through base complementation.
