MRNA vaccine for treating lung cancer and bone metastasis thereof as well as preparation method and application of mRNA vaccine
The invention discloses an mRNA (messenger Ribonucleic Acid) vaccine for treating lung cancer and bone metastasis thereof as well as a preparation method and application of the mRNA vaccine. Belongs to the technical field of nano-drug preparations and nucleic acid vaccines. The vaccine can realize co-entrapment of the TLR4 agonist monophosphoryl lipid A (mPLA) and mRNA (messenger ribonucleic acid). The particle size is 60-600nm, the Zeta potential is + 8 to + 37mV, the encapsulation efficiency is 60-93%, and the mRNA drug loading capacity is 7.5-20%. The mRNA vaccine constructed by the invention can be administered in a subcutaneous/muscle/nasal mucosa way, stimulate DCs cells to be mature, polarize M2 type macrophages into M1 type macrophages and activate the ADCC effect of NK cells, and multiple immune pathways synergistically play an anti-tumor treatment effect. Particularly, after bone metastasis of lung cancer occurs, the vaccine can effectively inhibit tumor growth, activate inherent immunity and adaptive response of a body by means of mPLA cooperating with mRNA immunotherapy, and cooperatively resist tumors..
