Construction method and application of oncolytic adenovirus carrying anti-ROR1/anti-CD3 antibody gene
The invention discloses a construction method and application of an oncolytic adenovirus carrying an anti-ROR1/anti-CD3 bispecific antibody gene. The construction method comprises the following steps: respectively constructing a plasmid pcDNA3.1-antiROR1 scFv and a plasmid pcDNA3.1-antiCD3 scFv; the method comprises the following steps: constructing a plasmid pCA13-ROR1BITE; the method comprises the following steps: constructing a plasmid pSD55-ROR1BITE; the method comprises the following steps: constructing pAd-ROR1BITE; and packaging is carried out, such that the oncolytic adenovirus ZD55-ROR1BITE carrying the anti-ROR1/anti-CD3 bispecific antibody gene is obtained. The construction method of the oncolytic adenovirus provides a new thought and method for research of liver cancer, and the oncolytic adenovirus constructed by adopting the method has a remarkable inhibition effect on liver cancer cells, so that a technical basis is provided for research of drugs for treating liver cancer..
