参苓白术散对溃疡性结肠炎大鼠NLRP3、NF-κB、MUC2、TFF3基因表达的影响 : = Influence of Shenling Baizhu Powder on mRNA expression of NLRP3,NF-κB,MUC2,TFF3 in rats of ulcerative colitis

目的观察参苓白术散对溃疡性结肠炎(UC)大鼠核转录因子κB(NF-κB)、炎症小体(NLRP3)、黏蛋白2(MUC2)、三叶因子3(TFF3)表达的影响,探讨该药治疗溃疡性结肠炎的作用机制。方法将60只雄性Wistar大鼠随机分为空白对照组、模型组、柳氮磺吡啶(SASP)组、参苓白术散高、中、低剂量组,每组10只。采用三硝基苯磺酸/乙醇灌肠液制备大鼠UC模型,SASP组给予SASP灌胃,参苓白术散各剂量组分别给予不同剂量参苓白术散灌胃,空白对照组、模型组灌服等量生理盐水,连续14 d。取材后采用real-time PCR检测各组大鼠结肠黏膜NLRP3、NF-κB、MUC2、TFF3 mRNA表达情况。结果参苓白术散可以明显下调NLRP3、NF-κB mRNA表达(P <0. 05),上调MUC2、TFF3 mRNA表达(P <0. 05),以中剂组效果最佳。结论参苓白术散可减轻肠黏膜炎症反应,促进结肠黏膜损伤修复,其机制与下调肠黏膜NLRP3、NF-κB mRNA表达,上调肠黏膜MUC2、TFF3 mRNA表达有关。.

Objective To study the influence of Shenling Baizhu Powder on mRNA expression of NACHT,LRR and PYD domains-containing protein 3 inflammasome(NLRP3),Nuclear Factor-Kappa B(NF-κB),mucoprotein(MUC2),Trefoil factor(TFF3) in rats of ulcerative colitis(UC),and to explore the mechanism of Shenling Baizhu Powder in treating ulcerative colitis. Methods Sixty male Wistar rats were randomly divided into normal control group,model group,sulfasalazine group,low-dose,middle-dose and high-dose Shenling Baizhu Powder groups,with 10 rats in each group. The UC rat model was built by trinitrobenzene sulfonic acid/ethanol enema. The Shenling Baizhu Powder groups were given Shenling Baizhu Powder by gavage; the sulfasalazine group was given sulfasalazine; the normal control group and model group were given saline with the same amount,for 14 days. The mRNA expression of NLRP3,NF-κB,MUC2,TFF3 in the intestinal mucosa was analyzed by real-time PCR. Results Shenling Baizhu Powder significantly down-regulated the mRNA expression of NLRP3 and NF-κB(P < 0. 05),and up-regulated the mRNA expression of MUC2 and TFF3(P< 0. 05). The middle-dose Shenling Baizhu Powder group had the most significant effect. Conclusion Shenling Baizhu Powder can reduce inflammatory reactions of intestinal mucosa and promote its repair. The mechanism may be associated with the down-regulation of the mRNA expression of NLRP3 and NF-κB and up-regulation of the mRNA expression of MUC2 and TFF3..
