基于互信息算法的抗前列腺癌药物重定位分析 : = Prostate cancer drugs repositioning based on mutual information
药物重定位研究不仅大大减少了新药开发的周期,还降低了经济成本,越来越受到医药界的重视。前列腺癌是世界上死亡率较高的癌症,近年来针对单个基因的研究不足以分析癌症的复杂发病机理。首先,应用R软件的互信息方法分析前列腺癌的基因表达数据并提取前列腺癌特征基因,得到从失联到关联状态下含有大量致病基因信息的特征基因。然后,通过cmap分析确定了几种治疗前列腺癌的药物。使用的基因表达数据来自TCGA,通过互信息分析算法发现了294条上调基因和362条下调基因。对这些特征基因进行cmap分析,发现了多种化合物对前列腺癌具有抑制作用,通过cmap负相关分数降序排列,得到了Thioridazine(硫利达嗪)、trichostatin A(曲古菌素A)等更可能对前列腺癌有抑制作用的药物。.
The study of Drug Repositioning not only greatly reduces the cycle of new drug development, but also reduces the economic cost. Prostate cancer is one of the cancers with high mortality in the world. In recent years,the single gene can not explain the pathogenesis of the cancers. In this paper, the mutual information method in R software was mainly used to analyze the gene expression data of prostate cancer and select the feature genes of prostate cancer, so as to obtain the feature genes which containing a large amount of pathogenic gene information from the state of disconnection to association. Then several drugs for prostate cancer were identified by cmap analysis. The gene expression data used in this paper were from TCGA, and 294 up genes and 362 down genes were found through mutual information analysis. Though the connectivity map analyzes these feature genes, then find that a variety of compounds has inhibitory effects on prostate cancer. Through connectivity map negative correlation fraction descending order, Thioridazine, trichostatin A and other drugs are more likely to have inhibitory effects on prostate cancer were obtained..
