手足口病假病毒中和抗体检测平台的建立和初步应用 : = Establishment and application of a surrogate neutralizing antibody detection platform based on HFMD pseudovirus
手足口病(hand,foot and mouth disease,HFMD)是婴幼儿常见的传染病,可并发重症及死亡。疫苗为防控HFMD的最有效措施。中和抗体是疫苗质控和评价的重要指标。为对疫苗研发提供工具,本课题组构建了HFMD主要病原肠道病毒A组71型(enterovirus A71,EV-A71)、柯萨奇病毒A组6型(coxsackievirus A6,CV-A6)、柯萨奇病毒A组16型(coxsackievirus A16,CV-A16)、柯萨奇病毒B组3型(coxsackievirus B3,CV-B3)和柯萨奇病毒B组5型(coxsackievirus B5,CV-B5)的假病毒,在此基础之上建立了肠道病毒假病毒中和抗体检测平台。本文简要概述了该平台的建立和初步应用结果。.
Hand,foot and mouth disease(HFMD)is a common infectious disease in infants.There is no effective antiviral treatment for severe HFMD cases.Therefore,vaccine is the most effective measure to prevent and control HFMD.Neutralizing antibody(NtAb)is an important indicator for the evaluation of vaccine efficacy and quality control.In order to provide tools for HFMD vaccine research and development,we first established pseudoviruses infection systems for HFMD associated viruses including enterovirus A71(EV-A71),coxsackievirus A6(CV-A6),coxsackievirus A16(CV-A16),coxsackievirus B3(CV-B3),and coxsackievirus B5(CV-B5),then a serial of novel pseudovirus expression firefly luciferase assays(PVLA)for the quantitative measurement of NtAb were developed based on these pseudoviruses.In this article,we describe the establishment and preliminary application results of this platform..
