口腔牙托水对大鼠细胞色素P450酶活性的影响 : = Effects of methyl methacrylate on CYP450 in rats

目的研究牙托水对大鼠CYP1A2、CYP2B6和CYP2C19酶活性的影响。方法 24只雄性SD大鼠,随机分为正常对照组、灌胃牙托水3 d组、灌胃牙托水10 d组。牙托水干预结束后,各组大鼠联合灌胃给予非那西丁10 mg/kg、安非他酮10 mg/kg和奥美拉唑10 mg/kg并测定探针药给予前及给药后的0.083 h、0.25 h、0.5 h、1 h、1.5 h、2 h、2.5 h、3 h、4 h、6 h、8 h、12 h和24 h的血药浓度。结果与正常对照组相比,10 d组大鼠奥美拉唑AUC下降;清除率CL/F显著性升高;平均驻留时间MRT下降;10 d组大鼠安非他酮表观分布体积V/F显著性降低,上述差异均有统计学意义(P<0.05);探针药的其余药代参数差异均无统计学意义(P>0.05)。结论牙托水可诱导肝CYP2C19酶活性,而对CYP1A2与CYP2B6无影响。.

Objective To elucidate the effects of methyl methacrylate on CYP1 A2,CYP2 B6 and CYP2 C19 in rats. Methods 24 male SD rats were randomly divided into 3 groups: Control group,MMA-3 d group and MMA-10 d group. All rats of three groups were administered intragastrically with 10 mg/kg phenacetin,10 mg/kg bupropion and 10 mg/kg omeprazole after administration of MMA for 3 or 10 consecutive days. Then,the blood samples were collected for analysis at 0,0. 083,0. 25,0. 5,1,1. 5,2,2. 5,3,4,6,8,12 and 24 h after administration. Results Compared with the normal control group,the omeprazole AUC decreased in the 10 d group; the clearance rate of CL/F increased significantly; the mean residence time of MRT decreased; the apparent distribution volume of bupropion( V/F) in the 10 d group significantly decreased,and the above differences were statistically significant( P < 0. 05). There were no statistical significance on the differences in the remaining pharmacokinetic parameters of the probe( P > 0. 05). Conclusion The results maybe indicated that MMA had the potential to induce rat hepatic CYP2 C19 activity,but not CYP1 A2 and CYP2 B6..
