肾动脉去交感神经支配术对慢性肾病大鼠肾功能及肾胺酶影响 : = Renal sympathetic denervation on kidney function and renalase in rats with chronic kidney disease

目的探讨肾动脉去交感神经支配术(RSD)对慢性肾病(CKD)大鼠肾功能及血压的作用,并观察其对肾胺酶蛋白表达的影响。方法将30只SD大鼠随机分为肾衰模型组(n=10)、RSD组(n=10)和正常对照组(n=10)。前两组以300 mg/(kg·d)腺嘌呤混悬液灌胃4周,8周时RSD组行RSD,分别在4、8周,RSD后4周测定血肌酐(Cr)值、尿素氮(Bun)值、尿量及24 h尿蛋白;8周及RSD术后4周测定血压;RSD术后4周蛋白质印迹法(Western blot)测定肾胺酶蛋白的表达。结果 RSD术后4周,肾衰模型组、RSD组Cr、Bun水平均明显高于正常对照组,RSD组Cr、Bun水平均低于肾衰模型组,差异均有统计学意义(P<0.05);肾衰模型组、RSD组24 h尿蛋白量均明显高于正常对照组,尿量均明显低于正常对照组,RSD组24 h尿蛋白明显低于肾衰模型组,差异均有统计学意义(P<0.01);RSD术后4周,肾衰模型组、RSD组收缩压(SBP)、舒张压(DBP)均高于正常对照组,RSD组SBP、DBP均低于肾衰模型组,差异均有统计学意义(P<0.05);RSD术后4周,RSD组肾胺酶的表达低于正常对照组,但高于肾衰模型组,差异均有统计学意义(P<0.05)。结论 RSD对CKD大鼠有延缓肾功能恶化、抑制尿蛋白增加、降低血压的作用,且能增加肾胺酶的表达。.

Objective To investigate the effect of renal sympathetic denervation(RSD) on renal function and blood pressure in rats with chronic kidney disease(CKD),and the expression of renalase. Methods A total of 30 SD rats were randomly divided into CRF group( n = 10),RSD group( n = 10) and control group( n = 10). Rats in the two former groups were fed with adenine 300 mg /( kg·d)for 4 weeks,RSD were accomplished after 8 weeks,blood serum creatinine(Cr),blood urea nitrogen(Bun),urine volume and urine albumin for 24 hours were measured in 4 weeks,8 weeks and 4 weeks after RSD,respectively; blood pressure were measured in 8 weeks and 4 weeks after RSD,respectively; renalase expression were detected by Western blot in 4 weeks after RSD. Results Four weeks after RSD,Cr and Bun in CRF and RSD groups were significantly higher than those in control group,Cr and Bun in RSD group was significantly lower than that in the CRF group( P < 0. 05); urine albumin for 24 hours in CRF and RSD groups were significantly higher than that in the control group,urine volume in CRF and RSD groups were significantly lower than that in the control group,urine albumin for 24 hours in RSD group was significantly lower than that in CRF group( P < 0. 01); after 4 weeks of RSD,systolic blood pressure(SBP),diastolic blood pressure(DBP) in CRF and RSD groups were higher than that in control group,SBP and DBP in RSD group were lower than those in the CRF group( P < 0. 05); 4 weeks after RSD,renalase expression in RSD group was lower than that in the control group,while higher than that in the CRF group( P < 0. 05). Conclusion Renal sympathetic denervation in rats with chronic kidney disease can delay renal function deterioration,restrain urine albumin,reduce blood pressure and increase renalase expression..
