鲍鱼腹足胶原蛋白的提取及性质研究 : = Isolation and characterization of collagen from the foot of Haliotis discus

测定了鲍鱼腹足的基本组成成分,对皱纹盘鲍酶促溶性胶原蛋白(PSC)的提取及相关性质进行了研究。结果表明,用酶法提取的鲍鱼腹足PSC得率为8.72%(以干基计)。圆二色谱分析表明,鲍鱼腹足PSC溶液在221和199nm分别有一正、负峰,是具有典型三螺旋结构的胶原蛋白特征圆二色谱峰型。鲍鱼腹足PSC等电点为pH 5.17;质量浓度0.3 mg/mL、pH 3.0~3.5的PSC溶液起泡性达110%,泡沫稳定性较差,乳化性好,为100%;但乳化稳定性一般,放置5h后,仅保持在50%左右。鲍鱼腹足PSC具有较高的Fe2+螯合活性,其IC50值为0.348mg/mL,当质量浓度为1.2mg/mL时Fe2+螯合能力可达91.62%。.

The component analysis of abalone adductor and extraction of pepsin-soluble collagen(PSC) from the foot of abalone(Haliotis discus) was performed,in which the yield of collagen was 8.72%(dry basis).The collagen solution shows the typical circular dichroism peak of collagen triple helix structure at 221 and 199 nm.The foaming of PSC was 110% at 0.03 mg/mL and pH 3.0-3.5 but the foam stability is poor.Its emulsion was 100% but the emulsion stability was 50% after 5 h.PSC has high Fe2+ chelating activity,IC50 value of which was 0.348 mg/mL.The chelating capacity was 91.62% when the concentration was 1.2 mg/mL..
