Activating natural killer (NK) cells through dendritic cell (DC) vaccination is highly beneficial, as NK cells can stimulate cytotoxic T cells (CTLs) and directly kill tumor cells. However, DCs matured with the standard protocol cannot secrete IL-12 and IL-15, crucial cytokines for NK cell activation. Electroporation of DCs with mRNA encoding a constitutively active form of IKKβ (caIKKβ) to activate the NF-κB pathway leads to the secretion of IL-12 and enhanced CTL activation. This study demonstrates that caIKKβ-DCs can efficiently activate NK cells, as evidenced by increased expression of CD54, CD69, and CD25 on NK cells, their ability to produce IFNγ, and their high lytic activity against target cells. Since caIKKβ-DCs still could not secrete IL-15, a chimeric IL-15/IL-15Rα construct was transfected into caIKKβ-DCs to further optimize NK cell activation, resulting in NK cells with even higher levels of activity..
Die Aktivierung von natürlichen Killerzellen (NK-Zellen) durch Dendritische Zellen (DCs) als Krebsimpfung ist vorteilhaft, da sie zytotoxische T-Zellen (CTLs) stimulieren und Tumorzellen direkt zerstören können. Jedoch können die standardmäßig verwendeten DCs die für die NK-Zellaktivierung zentralen Zytokine IL-12 und IL-15 nicht sezernieren. Die Elektroporation von DCs mit mRNA, die eine konstitutiv aktive Form von IKKβ (caIKKβ) codiert, aktiviert den NF-κB-Signalweg und ermöglicht eine IL-12-Produktion sowie effektivere T-Zellaktivierung. Diese Arbeit zeigt, dass caIKKβ-DCs auch NK-Zellen effizient aktivieren können. Die erhöhte Expression von CD54, CD69 und CD25 auf NK-Zellen sowie die Fähigkeit zur IFNγ-Produktion und hohe lytische Aktivität wurden nachgewiesen. Da caIKKβ-DCs jedoch kein IL-15 sezernieren konnten, wurde zusätzlich ein chimäres IL-15/IL-15Rα-Konstrukt in die DCs transfiziert.
